Difference in Osteoclast Responses to Tricalcium Phosphate in Culture Medium Supplemented with Zinc and to Zinc-Containing Tricalcium Phosphate

چکیده
Difference in osteoclast responses (i.e., apoptosis, actin ring formation) to tricalcium phosphate (TCP) in culture medium supplemented with zinc and to zinc-containing tricalcium phosphate (ZnTCP) was investigated in this study. On the TCP ceramic, an increase in zinc ion in the culture medium within the range between 0.3 and 6.8 ppm significantly induced an increase in osteoclast apoptosis and a decrease in actin ring formation. However, even a high level of magnesium ion up to 100 ppm in the culture medium or up to 6.8 mol% in the ceramic was unlikely to influence osteoclast activity. There was almost no significant difference in osteoclast apoptosis and actin ring formation between ZnTCP with 1 mol% zinc and magnesiumcontaining tricalcium phosphate with 1.5 mol% magnesium ceramic which have the same solubility and the same dissolution rate. It is suggested that only an increase in zinc level outside resorption lacuna has an inhibitory effect on osteoclast resorption and that an increase in zinc level inside resorption lacuna could not influence osteoclast activity.

Zinc in calcium phosphate mediates bone induction: in vitro and in vivo model.
Zinc-containing tricalcium phosphate (Zn-TCP) was synthesized to investigate the role of zinc in osteoblastogenesis, osteoclastogenesis and in vivo bone induction in an ectopic implantation model. Zinc ions were readily released in the culture medium. Zn-TCP with the highest zinc content enhanced the alkaline phosphatase activity of human bone marrow stromal cells and tartrate-resistant acid ph...

The aim of this study was to evaluate the therapeutic efficacy of biomimetic zinc-containing tricalcium phosphate (ZnTCP) produced by hydrothermally converting calcium carbonate exoskeletons from foraminifera, in the treatment of osteoporotic mice. X-Ray powder diffraction showed crystallographic structures matching JCPDS profile for tricalcium phosphate. Mass spectroscopy used to calculate tot...

Forty isolates of endophytic bacteria isolated from banana tree roots were assessed as to their capacity to solubilize phosphate in a solid culture medium supplemented with different inorganic and one organic source of phosphorus. The amount of phosphorus (P) in each liquid medium was quantified, and an indirect assessment of acid phosphatase activity was performed. All assays had a fully rando...

Zinc-containing β-tricalcium phosphate (ZnTCP) nano particles were injected into zinc-deficient rats to promote osteogenesis. Sprague-Dawley (SD) rats (4 weeks old, average weight of 70 g) were divided into four groups: Normal rats (not ovariectomized (OVX)), Control rats (OVX), and OVX rats injected with a suspension of ZnTCP nano particles or ZnSO(4). The ZnTCP contained 6.17% zinc. The suspe...
